Description |
mouse B cell hybridoma murine myeloma cells fused with C57Bl/6 spleen cells;
indicator cell line for IL-6 confirmed as mouse with IEF of MDH, NP, PEP B

Depositor |
Dr. E. Schmitt, Institute of Immunology, University of Mainz, Mainz, Germany
|
Morphology |
round cells, growing single or clustered in suspension
|
Culture_conditions |
90% RPMI 1640 + 5% FBS + 5% ConA spleen cell supernatant (or 10 ng/ml IL-6) +
2.5 x 10-5 M mercaptoethanol + 2 mM L-glutamine maintain at 0.1-0.3 x 106
cells/ml, optimal split ratio (daily) of 1:3, maximal split ratio of 1:5; for an IL-6 bioassay,
we recommend to initially use 1.25-2.5 x 104 cells/ml and 100 microl in a 96-well-plate (it
is advisable to determine the optimal cell density in various growth curves) at 37C
with 7.5% CO2 maximum density of 0.5-1.0 x 106 cells/ml

Viruses |
ELISA: reverse transcriptase positive; PCR: SMRV -

Properties |
bioassay for IL-6 (low cell densities are essential for reliable assays; try doubling cell
dilutions beginning with cell densities of 2.5 x 104 cells/ml; duration of assay is
